Effect of Fimbrial Abhesin in Esherichia coli on Host

Abstract
Bacterial adherence occurred by fimbrial adhesin is basic stage of infection. Our purpose in the experiments herein described is to confirm the role of fimbrial adhesin(or fimbrial antigen complex:FAC) in activities of PMNLs, epithelial
cells(Vero,
Caco
2) and the biochemical characteristics of fimbrial adhesin.
1. Highly fimbriated E. coli 43 had higher adherent capacity on PMNLs than poorly fimbriated E. coli 26. And E. coli 43 was more sensitive than E. coli 26 on pagocytosis and bactericidal capacity.
2. Purified fimbrial adhesin of E. coli had been prepared by gel filtration chromatography, and the molecular weight was measured about daltons with SDS-PAGE.
3. It was investigated that the effect of fimbrial adhesin of PMNs migration and PMNLs respiratory burst activity. Fimbrial adhesin stimulated significantly respiratory burst of PMNLs, and compared with phorbol ester and FMLP. It was suspected
that
fimbrial adhesin had the chemotactic activity and stimulated respiratory bust throughout the activation of protein kinase C.
4. The effect of fimbrial adhesin of adherence to Vero, Caco 2 cells was investigated. It was shown that E. coli 43, 1008 had excellent adherent capacity than E. coli 26, 437. Treatments of these strains with sub-MIC streptomycin showed the
markedly
reduced production of fimbrial adhesin and lower adherence to Vero, Caco 2 cells.
